ABSTRACT: DNA replication is a process fundamental to the faithful propagation of genetic information from one generation to the next. Using tools from the physical sciences, we are able to manipulate individual DNA molecules and observe how replication proteins copy the DNA. Observation of such complicated, biological processes at the single-molecule level provides us with 'molecular movies' that show details hitherto inaccessible by classical, ensemble-averaged biochemical experiments. We believe our work is a good example of how the bridging of disciplines enables researchers to address the important scientific questions of the 21st century.
